High Throughput Open Reading Frames Cloning and Heterologous Expression

We provide cloning and optimisation of conditions (temperature, IPTG concentration,
duration, protein solubility) for heterologous expression of Open Reading Frames (ORF)
from various organisms in E. coli.

Customer provides:

  • Genomic DNA
  • Complete sequence of each ORF that needs to be assayed

Our service includes:

  • Primer design and synthesis, cloning and sequencing of ORFs of interest
  • Trial expression of ORF in E.coli, protein solubility assay
  • Optimisation of conditions for heterologous expression according to the customer’s requirements:
        Temperature
        Inducer (e.g. IPTG) concentration
        Duration of expression
  • PAGE of samples obtained during optimisation.

Customer receives:

  • Report on ORF cloning and DNA sequence data analysed with standard software
  • Data on expression optimisation with PAGEs on protein yield and solubility
  • Optional: optimisation of buffer composition for protein extraction
  • ORF DNA in cloning and/or in expression vector.

Prices start from:

£100 per clone of individual ORF with confirmed DNA sequence,
£60 per trial expression,
£140 per optimisation of individual ORF expression.
The prices are based on a minimum of 50 ORFs per order.
