We offer monoclonal antibody (MAb) development in mice, with guaranteed success - three positive and stable clones against the antigen used in an ELISA. We have experience in raising antibodies against substances of low immunogenicity. All features of the antigen will be discussed before the start of a project. Customer will be regularly updated on the project’s progress.
We can predict success or failure of the project after mice immunisation. In case of a failure we provide complete report on the experiments, explain the reason for the failure and charge only for the first phase of the project – 10% of total cost.

 Customer provides:

  • Approximately 2-3 mg of protein antigen or 2 mg of conjugated peptide and 0.5 mg of free peptide for ELISA.
  • Information on the antigen nature and properties.

Customer receives:

  • 3 hybridoma clones, cells aliquots in 3 cryo vials per clone.
  • A certificate and protocol of the project.

 Standard Service:          

 Phase I (9 weeks)

  • Immunisation and boosts for 5 Balb/c mice with standard immunisation protocol.
  • Titer assay (ELISA) prior to selection of donor mice.

 Phase II (4 weeks)

  • Hybridoma fusion using splenocytes or lymph node cells and SP2/0 myeloma cells.
  • Screening of up to 2,000 hybridoma clones for positives with ELISA. Up to 10 highly positive clones will be subcloned, expanded, and 5 ml of hybridoma culture medium will be provided to customer for evaluation.

 Phase III (2 weeks)

  • Customer selects 3 clones that will be expanded for cryopreservation and sent to the customer.
  • We will keep those 3 selected hybridoma cells for next 6 months for free.

 Additional Services:        

  • Large scale antibody production in ascitic fluid or in stationary cell culture.
  • Peptide synthesis and conjugation to a carrier.
  • Additional clones for phases II and III.
  • Protein A or protein G affinity purification.
  • Antibody biotinylation.
  • Evaluation of suitable screening techniques other than ELISA.
