Description:

YB-GCPol DNA Polymerase* is a mix of thermostable enzymes, catalyses 5’→3’ synthesis of DNA. The enzyme has  3’→5’ proofreading, and possesses low 5’→3’ exonuclease activity. Enzyme mix is designed for processing DNA that has high GC-content or complex melting profile. Amplified DNA fragments can be used for subsequent cloning.  Properties of YB-GCPol.

Applications:

• PCR (primer extension, DNA amplification) with difficult templates - GC-rich, complex melting profile, “dirty”.
• Amplifying DNA for subsequent cloning

Features:

• Robust enzyme suitable for difficult cases
• Each 500 units of enzyme are supplemented with 0.25ml of  DNA-Softner – universal additive to difficult templates
• Enzyme can be supplemented with any 10x PCR buffer (see the List of PCR buffers) upon request

Unit Definition:

One unit of enzyme catalyses the incorporation of 10 nanomoles of deoxyribonucleotides into an acid-insoluble polynucleotide fraction in 30 minutes at 70 ºC.

Activity Assay:

50 mM Tris-Cl (pH8.8 at 25 ºC), 50 mM NaCl, 10 mM MgCl₂, 0.25 mg/ml activated calf thymus DNA, 0.2 mM each of dATP, dCTP, dGTP, and 0.05 mM of [³H] dTTP.
