This unique research service allows the customer to
estimate the functional efficacy of one or several siRNA targets and to choose
the best one for following in vivo
experiments. Our service includes quantitative functional validation and
efficacy measurements for any siRNA target sequence.
Customer provides:
The sequence of the siRNA to be tested (19 – 29 bases
corresponding to the
mRNA strand). Such sequence data can be obtained empirically using software
available online.
Our service includes:
- Oligonucleotide synthesis and cloning of the
indicated siRNA target sequence into a proprietary reporter plasmid.
Simultaneous preparation of the effector plasmid to express doublestranded siRNA directed to the same target carried by
the reporter
plasmids
- Effector plasmid and each of the reporter plasmids
will be cotransfected into HEK293 human cells in duplicates. Reporter
activity will be measured 48 h post transfection. Functional efficacy of the
given target will be calculated as the mean reporter silencing for upstream and
downstream target positions relating to the reporter coding sequence.
- Duration: 15-20 working days
